Colipan Dose, Use, Side Effect, Composition
Colipan is a drug containing hyoscine-n-butylbromide, the same composition as buscopan. It comes in tablets for adults and syrup for children. It is made in Nigeria by Nigerian-German chemicals plc.

Composition Of Colipan
Tablet
Each Sugar-coated tablet contains: 10 mg Hyoscine-N-butylbromide B.P.
Elixir
5 ml elixir contains: 5 mg Hyoscine-N-butylbromide B.P.
Properties
Colipan has a special site of action at the parasympathetic ganglia in the walls of the viscera. Because of this it exerts a specific antispasmodic action on the smooth muscle of the gastrointestinal, biliary and urinary tracts, this action being produced long before the side effects characteristic of atropine-like substances occur. Hence no side effects on the central nervous system or salivary glands are observed when Colipan is administered in therapeutic doses.

Indications And Use Of Colipan
Gastric and duodenal ulcers gastrointestinal spasm and hyper motility, pylorospasm in infants, post-operative vomiting, spastic constipation, biliary dyskinesia and spasm of the urinary tract, delayed relaxation of the lower uterine segment, dysmenorrhoea.
Colipan is also indicated in radiology and endoscopic procedures of gastrointestinal tract to reduce spasm.
Contra-Indications
There is no known contra-indication for the oral administration of Colipan. Colipan should not be administered parenterally in the following disorders. Glaucoma, hypertrophy of the prostate with a tendency towards retention of urine, mechanical stenosis in the region of the gastro-intestinal tract. tachycardia, megacolon.
Side Effects Of Colipan
No adverse effects characteristic of atropine are observed on the salivary glands or sweat secretion when Colipan is administered in the usual dose. A slight increase in pulse rate may occur when it is administered intravenously.
Parenteral administration, especially of large doses, may when it is administered occasionally cause a transient disturbance of accommodation, which recedes spontaneously. For these reasons, following parenteral administration, even when given as instructed, Colipan can affect vision and consequently reactions while driving or operating machinery.
Interactions
On parenteral administration of Colipan, tricyclic antidepressants, quinidine and amantadine can potentiate the anticholinergic effect of Colipan.
Special Precaution
Available but limited preclinical data has shown no hazard, but safety during pregnancy has not yet been established. The usual precautions regarding the use of drugs at this time, especially during the first trimester, should be observed. Safety during lactation has not yet been established.
Dosage Of Colipan
Unless otherwise prescribed by the Physician:
Oral
Adults and children over 6 years: 1-2sc tablets 3-5 times daily. The sugar-coated tablets should be swallowed whole with a little fluid.
Elixir
Adult and children over 6 years: 2-4 teaspoonfuls 3-5 times daily
Children over 1 year: 1-2 teaspoonfuls 3 times daily
Infants: 1 teaspoonful 3 times daily
(1 teaspoonful = 5 ml)
Parenteral
In acute attacks of colic, 1 ampoule of Colipan (20 mg) may be administered subcutaneously, intramuscularly or by slow intravenous injection several times daily.
Infants and young children: In severe cases, 1/4 of an ampoule 3 times daily.

Overdose
Symptoms
Sign of poisoning following acute overdose has not been observed in men. Single oral doses of up to 590 mg and quantities of the active substance of up to 1090 mg within 5 hours merely produced dry mouth, tachycardia, slight drowsiness, and transient visual disorders. Results from toxicity studies in animals after parenteral administration suggest the following possibilities: shock, cheyne-stokes respiration, respiratory paralysis, colonic spasms, paresis of the striated muscle, coma, paralytic ileus, cysto paralysis, and retention of urine.
Therapy
In the case of oral poisoning, gastric lavage with animal charcoal followed by magnesium sulfate (15%). Symptoms of Colipan overdosage respond to parasympathetics.
For patients with glaucoma, pilocarpine given locally boosts circulation with sympathomimetics. For coma, central analeptics. Neuroleptics for mental excitation.
In the case of respiratory paralysis, intubations, artificial respiration. In the case of orthostatic fall in blood pressure, it is sufficient for the patient to lie flat. Catheterisation in the case of urine retention.
