Difference Between Dry Granulation And Direct Compression

Tablet is defined as a compressed solid dosage form containing medicaments with or without excipients. They vary in shape and differ greatly in size and weight, depending on amount of medicinal substances and the intended mode of administration.

Direct compression vs dry granulation

Tablets are prepared by different methods. The choice depends on many factors. In this post, we shall see the difference and similarity between dry granulation and direct compression. Meanwhile, let us take a brief look at both method of tablet manufacturing.

Dry Granulation Method

Dry granulation or double compression, usually eliminates various steps, which involves slugging of the powder mass. The active ingredient, diluent and lubricant are blended together, to form the slug. Thus, the compressed slug is passed through the mesh or through the mill, and the remaining lubricant is added to the granulation, blended properly and compressed to form the tablets.

The steps are;

1. Obtain raw material

2. Weigh materials

3. Screening

4. Mixing of active ingredient, diluent and lubricant

5. Slugging is clumping of the mix

6. Milling the slug

7. Screening

8. Mixing

9. Compression

Direct Compression

Direct compression involves direct compressing the powdered material into tablets. Tablets prepared by the above method are subjected to a compression machine which may be single station or multiple stations.

The process are;

1. Raw material

2. Weighing

3. Screening

4. Mixing

5. Compression

Difference Between Dry Granulation And Direct Compression

1. In dry granulation, slugging may be used to form the granules but direct compression skip this process

2. Direct compression is adopted, if the drug constitutes a major portion of tablet (86-90) total weight but direct compression can be use with any weight of the drug

3. Steps in dry granulation are nine but direct compression uses five

4. Dry granulation utilises a suitable diluent which acts as a carrier or vehicle for the drug if the tablets contain 25% or less of drug substances but direct compression utilises diligent at all time

Similarity Between Dry Granulation And Direct Compression

1. Dry granulation and direct compression is used in case tablet ingredients are highly sensitive to moisture or elevated temperatures during drying
